Lynch will play Mother Superior, who runs the Catholic orphanage where the stooges grow up. Tony Award nominee Sean Hayes (Promises, Promises) recently signed on to play Larry. Chris Diamantopoulos will play Moe, and Will Sasso will play Curly.
Peter and Bobby Farrelly are set to direct the film.
Lynch won an Emmy Award, Golden Globe and a Screen Actors Guild Award for her work as Sue Sylvester on Fox's hit series "Glee." Lynch has also appeared Off-Broadway in Love, Loss, and What I Wore.
A release date for the film has not yet been announced.
